Foundayo is an oral, once-daily, investigational GLP-1 receptor pathway targeting therapy for chronic weight management in adults with obesity. Unlike injectable GLP-1 receptor agonists, orforglipron is a small-molecule, non-peptide option designed for convenient administration without food or water restrictions. Its mechanism focuses on appetite regulation, increased satiety, and reduced caloric intake, addressing key biological drivers of weight regain.
Orforglipron (Foundayo) is not intended for cosmetic weight loss and should not be used in combination with other GLP-1 receptor agonists. Safety considerations include a boxed warning for potential thyroid C-cell tumors (observed in animal studies), along with risks such as pancreatitis, gastrointestinal effects, hypoglycemia (especially with concomitant diabetes therapies), and dehydration. The therapy is contraindicated in patients with a history of medullary thyroid carcinoma or Multiple Endocrine Neoplasia syndrome type 2 (MEN 2). As a next-generation oral metabolic therapy, orforglipron represents a significant advancement in obesity pharmacotherapy, expanding access to effective weight management options through a non-injectable format.
