Unresectable hepatocellular carcinoma moved rapidly toward immunotherapy combinations, and the challenge was capturing CTLA-4 benefit without its cumulative toxicity. Imjudo (tremelimumab) was approved in a regimen built around that.
Given as a single priming dose alongside ongoing durvalumab, this CTLA-4 blocking antibody broadens the T-cell response once rather than exposing patients to repeated CTLA-4 dosing. The combination is approved for adults with unresectable hepatocellular carcinoma.
Imjudo also carries an indication in metastatic non-small cell lung cancer with durvalumab and chemotherapy. Its single-priming-dose design is the principal differentiator against ipilimumab-based regimens and a template other CTLA-4 combinations have followed.
